چکیده

THE FOLLOWING ANALYTES HAVE BEEN INVESTIGATED: urine albumin (u-albumin), plasma-transferrin (p-transferrin), p-haptoglobin, p-IgG, p-IgA, p-IgM, and p-orosomucoid. An unmodified commercial analytical system FIA Star (Tecator) with a two-channel injector (40 mul) was used. The prediluted plasma samples and antibodies are allowed to react for 33 s before the change in turbidity is measured as a Peak maximum at 405 nm. The optimal concentrations of calibrators and antibodies have been determined to secure antibody excess. Response time (i.e. delay between aspiration of a sample and presentation of the result in absorption units) is 75 s. Automatic print-out of the absorbance profile and movement of the sample rack further accounted for 21 s per sample, so the throughput is reduced to 75 determinations per 2 h. Results are available within an hour, compared to two-12 days with the present methods (electroimmunoassays). Parallel analyses with established methods/analysers show excellent agreement for u-albumin, p-transferrin and p-haptoglobin. For p-IgG, p-IgA and p-IgM the reaction time of 33 s is insufficient because their relative molecular masses (i.e. the size of the molecules) are so high, 150.000-971.000. Five minutes is a more adequate reaction time, which makes a serial analyser such as FIA Star unsuitable for larger workloads of samples of immunoglobulins. The plasma concentration of Orosomucoid is low, resulting in high sample blanks. It is therefore recommended that the reaction is followed kinetically if a serial analyser is used.

Profile and protective role of 61kDa and 47kDa antigens of Edwardsiella tarda in rohu (Labeo rohita)

Edwardsiella tarda is a versatile pathogen that could survive in different environmental conditions and infect economically important fish species. The whole cell antigenic proteins were extracted from E. tarda and assessed via Western blot using anti E. trada rohu and anti E. trada rabbit serum. Two strong reacted proteins with antibodies were recovered viz 61 kDa and 47 kDa from SDS PAGE gels...

CHROMOSOMAL PROTEINS IN PULMONARY ALVEOLAR MACROPHAGES

In this study the nature of chromosomal proteins, his tones and nonhistone in resident alveolar macrophages was investigated in comparison to peritoneal neutrophils and calf thymus proteins. Cells were obtained by lavaging the lung and after purity determination they were subjected to fractional extraction procedures. Proteins were then analysed on SDS polyacrylamide gels and densitometric...

NOTES & UNIQUE PHENOMENA Using a Nitrate Specific Ion Electrode to Determine Stalk Nitrate-Nitrogen Concentration

The end-of-season stalk NO, test has been used to determine N sufficiency in corn (Zea mays L.). Nitrate concentration is commonly determined with flow-injection analysis (FIA), which is accurate but uses hazardous chemicals and is time-consuming. Use of a simpler method of NO, determination, such as the NO, specific ion electrode (SIE), may save time and costs, and reduce hazards. The objectiv...

Investigation of Network of Proteins Effective in Esophageal, Gastric and Colon Cancers and Determination of Key Common Proteins Using In Silico Method

Introduction: The prevalence of gastrointestinal cancers, especially gastric and colorectal cancers, is increasing. Gastric cancer is the most common gastrointestinal cancer in Iran and the age of esophageal and colon cancers onset has decreased in Iran. Therefore, it is important to study the factors and proteins involved in these cancers. Use of computational tools is an effective way to stud...

Determination of Superficial Clefts on Fragment of Antigen Binding in Human Immunoglobulin G by Computational Immunology

Background: Immunoglobulins (Igs) are protective glycoproteins specifically identify and eradicate microbes. Fragment of antigen binding (Fab) is a portion of antibody which binds to antigen and consists of one variable and one constant domain of one heavy and one light chain. Idiotypes, epitopes situated on Igs variable region, could be exploited to monitor and target malignant B cells and are...
